PE-22-28 (Mini-Spadin): High-Affinity TREK-1 Inhibition, Plasticity-Associated Signaling, and Sortilin-Propeptide Pharmacology in Preclinical Models

Abstract PE2228, also called minispadin, is a synthetic sevenaminoacid peptide with the sequence GlyValSerTrpGlyLeuArg (GVSWGLR). It is derived from residues 22–28 of the 44residue propeptide released during sortilin maturation and was developed as a shortened functional analogue of spadin, the longer PE12–28 sortilinpropeptide derivative. In electrophysiology experiments using human TREK1/KCNK2expressing HEK cells, PE2228 displayed high apparent potency for inhibition of the twoporedomain potassium channel TREK1, with a reported IC₅₀ of approximately 0.12 nM in the authors’ assay. The same study reported experimental selectivity across a limited K2P and hERG channel panel, together with behavioral and neuroplasticityassociated findings in mice [1]. This review summarizes PE2228 as a preclinical ionchannel research tool. It distinguishes direct electrophysiology, mousebehavior, neurogenesismarker, and cellculture findings from unsupported human claims. It also separates the parent peptide from modified G/APE2228 and biotinylated analogues, whose stability and duration observations cannot automatically be assigned to unmodified PE2228. No published human pharmacokinetic, doseranging, efficacy, or longterm safety program was identified in the reviewed literature. Accordingly, PE2228 should be considered an investigational research compound rather than a treatment for depression, cognition, stroke, diabetes, or any other condition. 1. Molecular Identity and Development Lineage 1.1 Exact Sequence and Chemical Profile PE2228 is a linear heptapeptide: HGlyValSerTrpGlyLeuArgOH, abbreviated GVSWGLR. PubChem lists the molecule as CID 165437303, with molecular formula C₃₅H₅₅N₁₁O₉, molecular weight 773.9 g/mol, and CAS number 1801959125 [2]. Structurally, the sequence combines an aromatic tryptophan residue, an arginine guanidinium group, a serine hydroxyl group, two glycine residues that add backbone flexibility, and the hydrophobic valine and leucine side chains. This mixed physicochemical architecture helps explain why a sevenresidue fragment can retain specific channel activity despite being substantially shorter than its parent sequence. 1.2 From Sortilin Propeptide to Spadin to MiniSpadin Sortilin, also known as neurotensin receptor3 (NTSR3), is synthesized as prosortilin and maturates through furinmediated release of a 44residue Nterminal propeptide. The longer synthetic spadin sequence corresponds to residues 12–28 of this propeptide. In the original spadin work, the PE12–28 sequence was designed to preserve a segment thought to maintain functional conformational properties while retaining activity against TREK1 [3]. In later structure–activity work, the researchers screened truncated derivatives and identified PE2228 as the shortest active sevenresidue core sequence, leading to the minispadin designation [1]. This nomenclature matters. PE2228 is not a fulllength endogenous hormone, and it is not simply a naturally circulating sevenresidue fragment whose human concentration and systemic role are established. It is a synthetic, sequencedefined research derivative designed from the sortilin propeptide / spadin pharmacology program. The molecular target and downstream biology should therefore be interpreted according to the evidence generated with the specific parent compound, unmodified minispadin, or individual modified analogues. 2. TREK1 Channel Biology and Target Rationale 2.1 TREK1/KCNK2 as a TwoPoreDomain Potassium Channel TREK1 is encoded by KCNK2 and belongs to the K2P family of twoporedomain potassium channels. These channels help set resting membrane potential and regulate cellular excitability. TREK1 activity is influenced by mechanical stretch, membrane lipids, temperature, pH, polyunsaturated fatty acids, and volatile anesthetics. It is expressed in central nervous system circuits and has also been implicated in peripheral physiology, including pain, anesthesia, and cardiovascular biology [4]. Its broad distribution makes it attractive for mechanistic research but also means that target engagement in a cell line cannot by itself establish a benign wholeorganism safety profile. 2.2 Sortilin–TREK1 Interaction and the Spadin Concept Foundational studies reported that sortilin and TREK1 can interact in neuronal systems and that sortilin expression affects TREK1 delivery to the plasma membrane. The same research program found that spadin bound to and inhibited TREK1 in cellular and neuronal electrophysiology experiments, while animal studies linked TREK1 inhibition to altered serotonergic activity and depressionlike behavior readouts [3]. These observations provided the conceptual basis for designing smaller sortilinderived TREK1 modulators, including PE2228. “From the study of spadin blood degradation products, we designed a 7 aminoacid peptide, PE 2228.” — Djillani et al., Frontiers in Pharmacology, 2017 [1] 3.
